KASP genotyping protocols for your qPCR instrument
Performing KASP™ genotyping in your own lab is easy. You need your DNA samples, your ordered KASP assays, universal KASP-TF Master Mix from LGC and a FRET-capable plate reader or a qPCR instrument.
It is straightforward to perform KASP™ genotyping in your own laboratory. Once you have your DNA samples, and have ordered your SNP-specific KASP assays and universal KASP-TF Master Mix from Biosearch Technologies, all that is required to read the fluorescent signal from completed KASP reactions is a FRET-capable plate reader or a qPCR instrument.
The table below outlines the optimal ROX level of KASP-TF Master Mix for the most commonly used qPCR instruments. Where applicable, a manual for running KASP on the specific qPCR instrument can also be downloaded.
KASP-TF Master Mixes are formulated without Triton™ X-100. For FRET-capable plate readers, Biosearch Technologies recommends initial trials with Standard ROX KASP-TF Master Mix. Run one of our free-of-charge trial kits on your qPCR instrument before running your own DNA samples and KASP assays.
Please note that KASP is currently available for endpoint genotyping only.

*For these instruments, we recommend Standard ROX KASP Master mix although the instruments will work with both low and high ROX mixes. These instruments can read ROX but the instrument-specific software does not use ROX values for normalisation. Running KASP with standard ROX KASP Master mix gives the user the option to export data to alternative software to perform normalisation if required.
