Thymine Linked BHQ Dyes
The BHQ dye is linked through a thymine rather than through a phosphate to allow more flexibility in designing assays.
LGC Biosearch is expanding our thymine linked BHQ® (Black Hole Quencher®) product line of synthesis reagents to include the thymine linked BHQ-1 and BHQ-2 CPG’s (Controlled Pore Glass) for 3’ labeling. The BHQ dye is linked through a thymine rather than through a phosphate to allow more flexibility in designing assays.
BHQ T-Linker CPG is used to add a non-fluorescent quencher at the 3' end of an oligonucleotide. These supports have a glycolate linkage to the CPG, which allows for rapid cleavage of the oligo and compatibility with base-sensitive fluorophores. Once cleaved from the support, the 3’ BHQ T-linked modification has a hydroxypropyl ester, which prevents polymerase extension and ligation at the 3’ end of the probe.
The thymine linked BHQ dyes allow flexibility with your assay designs, enabling alternative chromophore linkage arrangements. A thymine base which can participate in hybridization can be incorporated into the oligo without the disruption of the phosphate backbone allowing expanded possibilities for probe design.
The BHQ-1 quencher has maximal absorption in the 480 to 580 nm range, which provides excellent quenching of fluorophores emitting in this range, such as FAM, TET, JOE, HEX, CAL Fluor® Gold 540, and CAL Fluor Orange 560 dyes.
The BHQ-2 quencher has maximal absorption in the 560 to 670 nm range, which provides excellent quenching of fluorophores emitting in this range, such as TAMRA, Cy3, Quasar® 570, Quasar 670, Cy5, CAL Fluor Red 590 and CAL Fluor Red 610 dyes.
The T-linked BHQ CPG supports are available on 500Å CPG for synthesis of oligos up to 50-mers in length.
