What is your recommended method for reconstituting and storing oligonucleotides?
To reconstitute dry oligonucleotides:
To prepare a 100 µM stock of reconstituted probe: Multiply the total nmol value provided on the Certificate of Analysis, by 10. The resulting number will be the volume of diluent (in microliters) to add to your probe. Once resuspended, the probe will be 100 µM and represents a stock solution.
For example: If the delivered amount was 20 nmol, then:
(20) x (10) = 200 microliters is needed to reconstitute the oligo to 100 µM.
- We recommend a short centrifugation of the product tube to ensure the oligonucleotides pellet is at the bottom.
- Resuspend the product in an appropriate volume of solution such as TE buffer (10 mM Tris, 1mM EDTA, pH 8), to achieve a stock concentration of 10 µM or more, ideally 100 µM.
- Allow the oligo to rehydrate for several minutes at room temperature and vortex for 15 seconds.
- Product should be exposed to a minimum of freeze/thaw cycles to avoid oligo degradation. We recommend aliquoting the stock solution into multiple aliquots for storage at -20 °C or cooler, in the dark. When stored in this manner, we anticipate product stability for one year from delivery.
