Universal controls for Stellaris® RNA FISH are convenient and easy-to-use positive controls for smooth adoption of the Stellaris technology. The Stellaris universal controls hybridise to the polyA tail of both immature and mature mRNAs to produce a well-known fluorescence pattern consistent with their cytoplasmic and nuclear distribution.

Key Benefits

  • A reliable and inexpensive positive control for validation and optimization of RNA FISH experiment setup

  • Hybridise to the polyA tail of both immature and mature RNAs

  • Produce a well-known fluorescence pattern consistent with broad cytoplasmic distribution of mature RNAs, lncRNAs, and pre-RNAs in nuclear speckles

*To improve your overall experience with your Stellaris product we are recommending ATTO647N T-30 controls in lieu of Quasar 670. Additionally, all Quasar 670 probe sets are now offered with ATTO647N T30 universal controls.

ATTO 647N has a similar ex/em spectra (646/664 nm) to Quasar 670 (647/670 nm) and can be detected in the same channel.

ATTO 647N belongs to a new generation of fluorescent labels for the red spectral region with the following characteristic features:

  • Strong absorption
  • High fluorescence quantum yield
  • High thermal and photo-stability
  • Exceptionally high stability towards atmospheric ozone

Figure 1. Stellaris universal control.
The T30 single labeled oligo is compatible with the Stellaris protocols. The T30-oligo reveals the intracellular polyA distribution in a “one-pot shot” for easy cell boundary identification for single molecule RNA detection and counting. Shown here is a combination-experiment with mouse C2C12 cells. The polyA RNA was probed with a Quasar 570 labeled T30 oligo (left and right panel; green cytoplasm and nculear speckles), mouse Tbp mRNAs with a probe set labeled with Quasar 670 (middle and right panel; red cytoplasmic spots), and nuclear DNA stained with DAPI (middle and right panel; blue nucleus).
